BLAST Hit for Human Adenovirus 26
Methods
Sequencing was performed as described previously. Briefly, DNA was isolated using a CTAB/Chloroform/SPRI method. Sequence libraries were constructed with Watchmaker Genomics Illumina Fragment library kit. Libraries were sequenced on an Illumina HiSeq4000. Reads were trimmed with TrimGalore and assembled with MegaHit. The largest contig was annotated with SnapGene.
Discussion
The Spike sequence in the J&J Ad26.COVS.2 vaccine has a different codon optimization than Pfizer BNT162b2. This can impact Primer design for qPCR assays attempting to Target multiple vaccines. MGC Spike qPCR and RT-qPCR primers described previously are a 100% match. The MGC Ori Primers only target the Bacterial Origin in Moderna and Pfizer monovalent and bivalent mRNA vaccines. The Ori qPCR/RT-qPCR assay does not hit this Adenovirus vector.
